In one challenging project, multiple providers had declined an order involving a target gene that was the reverse complement of an existing vector sequence. Once inserted, the two approximately 1,000 bp complementary regions would form a seamless hairpin structure, making stable cloning particularly challenging.

How did Synbio Technologies solve it?
Rather than relying on conventional homologous recombination, our team developed a tailored workflow combining:
- Vector modification and restriction enzyme–ligation cloning
- Specialized competent cells for hairpin-containing constructs
- Careful clone screening and restriction digest analysis
- Third-generation sequencing to verify the full-length construct, junctions, and hairpin structure
The result: a successful delivery of a construct that others had been unable to take on.

Why Does It Matter?
Reliable synthesis and cloning of complex DNA structures can support downstream research in RNA and DNA structural biology, gene regulation, functional genomics, and the development of specialized genetic constructs.
